Automating the CGH/CNV Workflow with the Bravo Automated Liquid Handling PlatformA new method was developed to automate CGH/CNV sample fragmentation, labeling, purification, and pre-hybridization setup in a 96-well format by using the Agilent Bravo Automated Liquid Handling Platform. Protocols for both the standard enzymatic (Genomic DNA Enzymatic Labeling Kit) and ULS (Genomic DNA High-Throughput ULS Labeling Kit) methodologies have been developed and validated with VWorks Automation Control software. The new workflow, compatible with both SurePrint HD and G3 microarrays, enables superior performance across the entire Agilent CGH/ CNV portfolio. Complete Automation of Quantitative PCR Setup on the Agilent Bravo Automated Liquid Handling PlatformThe Agilent Bravo Automated Liquid Handling Platform has been shown to be an effective way to automate setup of a variety of quantitative polymerase chain reaction (QPCR) assays. QPCR reactions to examine a variety of housekeeping genes from cDNAs derived from different human tissues were set up on the Bravo Automated Liquid Handling Platform and by hand. Dilutions performed by each method showed similar slopes and good linearity and targets amplified by either method also gave similar Ct values. When examining cDNAs from fetal and adult lung, differential expression was observed in the B2M whereas GUSB and TBP remained similar. The Bravo Automated Liquid Handling Platform should be useful to scientists interested in speeding up setup while maintaining reliability when performing QPCR studies.> Download Application Note

Complete Automation of the Stratagene StrataPrep 96 PCR Purification Kit with the Agilent Bravo Automated Liquid Handling Platform and Agilent Automated CentrifugeA new method was developed to automate PCR purification in a 96-well format using the Bravo Automated Liquid Handling Platform, Automated Centrifuge and StrataPrep 96 PCR Purification Kit from Agilent Technologies (Automation Solutions and Stratagene divisions, respectively). Current PCR purification protocols require extensive manual processing including repetitive pipetting and vacuum filtration steps. Using Agilent’s VWorks Automation Control software, a protocol was developed for the Bravo Platform to aspirate, dispense, mix, and transfer sample from a source 96-well PCR plate to the binding and collection plates of the StrataPrep 96 kit as well as to automate sample wash steps using an accessory vacuum filtration unit on the Bravo Platform. Sample recovery was accomplished using the Agilent Automated Centrifuge. DNA recovery quality was assessed on the Agilent BioAnalyzer 2100.> Download Brochure

Automation of Agencourt AMPure Purification Kit for the Purification of Next-Generation Sequencing Sample Preparation Reactions on BravoA new method was developed to automate library construction for the Illumina sequencing platform using the Agilent Bravo Automated Liquid Handling Platform and the Agencourt Ampure PCR purification kit. Current Next-Generation sequencing workflows require extensive manual processing steps including repetitive pipetting and the use of individual centrifugation columns for cleanups of enzymatic steps. Using the Agilent VWorks Automation Control software, a protocol was developed for the Bravo platform to aspirate and dispense enzyme master-mixes, and perform cleanups of these reactions through the transfer of the Agencourt AMPure reagent to the reaction, transfer of the microtiter plate to a magnet using the integrated gripper, and subsequent sample wash and elution steps. Sample quality and quantity were assessed using the Agilent Bioanalyzer 2100 and Illumina Genome Analyzer IIX.> Download Application Note

Using Agilent’s VWorks Automation Control software, a script was written for the Bravo Platform to aspirate, dispense, mix, and transfer sample from a source 96-well tissue culture plate to the binding and collection plates of the Absolutely RNA 96 kit as well as to automate sample wash steps using an accessory vacuum filtration unit on the Bravo Platform.
